Calcium ion influx is a characteristic of myocardial excitation-contraction coupling. Cardiac organoids have a pacemaker site similar to the sinoatrial node, driving the contraction of the entire three-dimensional sphere. After X-ray radiation, changes in myofibrils were observed under an electron microscope, prompting us to investigate the calcium signals in cardiac organoids. Using high-resolution fluorescence mapping technology, we evaluated the calcium transient (calcium transients) changes in cardiac organoids, studying the alterations in indicators such as the speed, dispersion, amplitude, frequency, and repolarization time of the calcium signal activity. The results showed that UCMSCs-sEVs treatment reversed the abnormal calcium handling events caused by radiation damage.
