To detect the influence of this pathway on atrial electrical activity and whether the above pathway works through sympathetic and parasympathetic nerves, a Electrical cardiac electrical mapping technique was employed to detect field signals in isolated hearts after Langendorff perfusion. Electrical mapping of cardiac electrophysiology demonstrated that the induced rate and duration of AF in depressed rats were significantly increased, and decreased in the Man1a2 knockdown group, respectively (Figure 7G and H,). At the same time, we also noticed that the AERP of isolated rat hearts in depression group did not decrease significantly (Figure 7I,). Particularly, the electrical conduction dispersion of the left atrium in CUMS-depressed rats increased markedly, and it decreased abnormally after Man1a2 knockdown (Figure 7J–N, Figure S8 and S9). Among them, the left ventricular activation time under stimulation also showed a different trend, but it may not show the therapeutic effect of Man1a2 knockdown due to the limitation of sample size (Figure 7L). This means that Man1a2 can be a target to reduce the occurrence of AF in the depression model. Additionally, the right atrium did not increase the dispersion of impulse conduction and the specific underlying reasons deserve further discussion

Fig. 7: Knockdown of Man1a2 reverses autophagy and NRAP in depression rats 2 (G, H) The isolated heart of depressed rats still showed high AF-inducing rate and duration, which could be improved by knocking down Man1a2 (n = 3). (I) There was no significant difference in AERP among the groups in vitro electrical mapping. (J, K) The dispersion of impulse conduction in the left atrium of the heart of the depressed rat increases, which can be improved by knocking down Man1a2 (n = 3). (L) The active time of depressed rats after left atrial stimulation was significantly prolonged (n = 3). (M) The picture is the field signal detected by the Electrical cardiac electrophysiological mapping technique after Langendorff perfusion in the isolated rat heart. (N) The dispersion phase diagrams of the left atrial impulse conduction in rats is shown. Depressed rats showed that the dispersion of impulse conduction increased significantly, and the dispersion of impulse conduction reversed significantly after Man1a2 knockdown (n = 3).
